Beta-alanine is a non-essential amino acid that, when supplemented chronically, increases intramuscular carnosine — the primary intracellular buffer against the hydrogen ion accumulation that causes fatigue during high-intensity exercise. Unlike sodium bicarbonate, which operates extracellularly in the blood, carnosine buffers directly inside the muscle fibre. This distinction determines which events and intensities benefit most from each supplement.
The evidence base for beta-alanine is unusually strong for a sports supplement: 40+ RCTs, multiple independent meta-analyses, and well-characterised dose-response relationships across sport types. It is one of the five supplements with a strong peer-reviewed evidence base for endurance performance, alongside caffeine, creatine, nitrates, and sodium bicarbonate.
The Mechanism: Raising Muscle Carnosine
Carnosine (beta-alanyl-L-histidine) is synthesised in skeletal muscle from beta-alanine and histidine. In most athletes, intramuscular beta-alanine availability — not histidine — is the rate-limiting step for carnosine synthesis. Supplementing oral beta-alanine raises plasma beta-alanine, driving increased muscle carnosine synthesis over 4–16 weeks. Which Events and Intensities Benefit
Intramuscular carnosine buffering is most critical at intensities above the lactate threshold — Zone 4 and above — where significant intracellular acidosis develops:
- 1–4 minute efforts: Largest effect sizes; 800m–1500m running and 2000m rowing. 1–3% performance improvement in meta-analyses.
- 4–10 minute efforts: Moderate evidence for 5km running and 4000m cycling pursuit. ~1–2% improvement.
- Sprint-finish capacity: Improved final kilometre pace and sprint capability; relevant for road racing where a finishing surge determines the result.
- Interval training quality: Reduces performance decrement across repeated hard intervals; improves training session quality during high-intensity blocks.
- Steady-state events above 60 min at aerobic intensity: Limited evidence for benefit — intramuscular acidosis is not the primary fatigue mechanism at these intensities.
Dosing Protocol
Standard maintenance dose: 3.2–6.4g per day, split into smaller doses (0.8–1.6g each) to minimise paraesthesia. Higher single doses increase tingling without accelerating loading proportionally.
- Loading phase: 6.4g/day in 4 divided doses (1.6g × 4) for 4 weeks; then reduce to maintenance
- Maintenance: 3.2g/day in 2 divided doses, or 4.8g/day if targeting peak levels
- With meals: Taking beta-alanine with carbohydrate-containing meals increases muscle uptake by ~40% via insulin-facilitated transport
- Sustained-release formulations: Reduce paraesthesia at equivalent daily doses without compromising loading efficacy
Paraesthesia: The Tingling Sensation
The characteristic tingling or flushing sensation experienced 15–30 minutes after ingestion is a benign pharmacological effect mediated by MRGPRD receptors in sensory neurons — not an indication of toxicity. It subsides within 60–90 minutes. Mitigation: split doses to a maximum of 1.6g per dose, take with food, or use sustained-release capsules. The tingling does not indicate a problem and does not affect ergogenic efficacy.
Stacking and Periodization
Beta-alanine operates via chronic loading — there is no benefit to timing doses acutely around workouts beyond consistent daily intake.
